This means that every time you visit this website you will need to enable or disable cookies again.The ultimate objective of this proposal is to develop a genetic transformation system in the freshwater planarian, Dugesia japonica. In the final year of the present grant period, we have cloned homologues of a zebrafish cDNA encoding D. japonica dorsal head antigen (DjDHA), a previously identified zebrafish gene that is expressed in the olfactory region of the ventral head. Transgenic D. japonica planarians harboring the DjDHA construct in their cells will be able to test whether this antigen is an effective, transgene delivery vehicle. In the first specific aim of this proposal, we will use quantitative PCR to determine whether DjDHA is broadly expressed in the brain, neural tissue, and/or olfactory epithelium of normal planarians. The second specific aim will determine whether overexpression of the DjDHA construct in planarians results in a transgene phenotype. We will conduct a subtractive hybridization experiment to compare gene expression in transgenic and non-transgenic planarians and will confirm expression by western blotting.
